Aims: To evaluate if Lemon Extract Reduces Angiotensin Converting Enzyme (ACE) Expression and Activity and Increases Insulin Sensitivity and Lipolysis in Mouse Adipocytes

Methods: Reagents, Cell Culture, Preparation of Lemon Extract (LE), ACE Inhibitory Activity Assay and others.

Results: Lemon Extract Decreases ACE Gene and Protein Expression as Well as ACE Enzymatic Activity. A significant (p < 0.002) and robust (12.00 +- 0.05-fold) increase in ACE gene expression was observed during 3T3L1 adipogenesis (Figure 1A). Incubation of mature differentiated 3T3L1 adipocytes with total LE (100 µg/mL) resulted in a significant reduction in ACE gene expression, which was reduced by 80.0% +- 0.5% after 10 h. Differentiated 3T3-L1 cells were treated with LE doses of 50, 100 and 500 µg/mL. There was no significant change in ACE protein expression within the 3 doses used, likely because 50 µg was already the maximum effect on the protein expression (Figure 1C). However, there was a 40.0-55.0 +- 0.4% (p = 0.01) reduction in protein expression compared with untreated cells. This was tightly correlated with ACE reduced activity (15% up to 87%) with increasing concentrations of LE. ACE activity was quantified by measuring the conversion of ACE substrate Abz-GLY-Phe(NO2)-Pro into Abz-GLY product.

Conclusions: The LE-dependent fat breakdown properties observed in mature adipocytes rationalizes its use as dietary intervention. The link between LE and ACE expression levels and LE as an ACE inhibitor also provides a mechanism based upon which LE is not only useful in the management of obesity but could also be beneficial in the management of cardiovascular diseases.

DMI Analysis of the Above Clinical Trial

The study aimed to evaluate the effects of lemon extract (LE) on angiotensin-converting enzyme (ACE) expression and activity, as well as its potential to enhance insulin sensitivity and lipolysis in mouse adipocytes, particularly 3T3-L1 cells. The methodology encompassed various techniques, including cell culture, preparation of lemon extract and an ACE inhibitory activity assay.

Results demonstrated that lemon extract significantly reduced both ACE gene and protein expression, alongside ACE enzymatic activity. Notably, during the adipogenesis of 3T3-L1 cells, ACE gene expression markedly increased by 12.00 +- 0.05-fold (p < 0.002), indicating the enzyme's potential role in adipocyte differentiation. Subsequent treatment of mature differentiated 3T3-L1 adipocytes with total lemon extract at a concentration of 100 µg/mL led to an impressive reduction in ACE gene expression by 80.0% +- 0.5% after 10 hours, suggesting a strong inhibitory effect of LE on ACE.

The analysis further revealed that while there was no significant change in ACE protein expression across doses of 50, 100 and 500 µg/mL, a notable 40.0-55.0 +- 0.4% reduction (p = 0.01) was observed compared to untreated cells, reinforcing the notion that lower doses might already elicit maximum effects. This reduction in protein expression correlated closely with decreased ACE activity, which ranged from 15% to 87% with increasing lemon extract concentrations.

Overall, the findings suggest that lemon extract's ability to influence ACE expression and activity not only supports its role as an effective dietary intervention for enhancing fat breakdown in adipocytes but also highlights its potential benefits for managing obesity and related cardiovascular conditions. This research provides a mechanistic basis for utilizing lemon extract in dietary strategies aimed at improving metabolic health and preventing cardiovascular diseases.
